Tip: vials are physical glass vials, usually 3 mL or 5 mL. You can’t add more bacteriostatic water than the vial holds, so your water amount is capped by the vial size.
Reconstitution means mixing a lyophilized (freeze-dried) peptide with bacteriostatic water so it can be drawn into a syringe. The math is simple: concentration (mg/mL) = vial amount (mg) ÷ water added (mL). To find how much to inject, divide your desired dose by the concentration to get the volume, then multiply by 100 to get insulin-syringe units.
A 5 mg vial with 2 mL of water is 2.5 mg/mL, or 2500 mcg/mL. For a 250 mcg dose: 250 ÷ 2500 = 0.1 mL = 10 units on a U-100 syringe.

Concentration equals the vial amount in milligrams divided by the bacteriostatic water added in milliliters. Dose volume equals your desired dose divided by that concentration. On a U-100 insulin syringe, units equal the dose volume in milliliters times 100.
There is no single correct amount. More water makes each unit easier to measure but dilutes the dose. Many people add 1 to 3 mL to a vial and let the math set the dose volume. Choose an amount that makes your target dose easy to draw.
On a U-100 insulin syringe, 100 units equals 1 mL, so 1 unit equals 0.01 mL.
