TUDCA

Mechanism

A taurine-conjugated, hydrophilic bile acid (the taurine conjugate of UDCA) that acts as a chemical chaperone reducing endoplasmic reticulum (ER) stress and protein misfolding, stabilizes mitochondria, and has anti-apoptotic and cytoprotective effects, including in hepatocytes.

Half-life

Not well established in humans; bile acids undergo enterohepatic recirculation, complicating a simple half-life estimate (uncertain).

Regulatory status

Not FDA-approved as a drug in the US; sold as a dietary supplement. (The parent compound UDCA/ursodiol IS FDA-approved for certain cholestatic liver diseases, but that is a distinct molecule.) TUDCA is approved/used as a pharmaceutical for cholestasis in some other countries (e.g., parts of Europe and Asia), but not by the FDA.

Safety

Generally well tolerated in studies; most common reported effects are mild GI symptoms (e.g., diarrhea). Long-term human safety data at supplement doses are limited; quality varies across unregulated supplement products.

Evidence

Some human trials (cholestatic liver disease, metabolic/ER-stress conditions) plus substantial preclinical data; evidence for many marketed supplement claims is still emerging.
